SMITE 2 offers a variety of gamemodes, each with its own unique map, playstyle, rules, and objectives. While each mode provides a different experience, they all share common elements such as minions and structures that influence the flow of gameplay. New players can queue for Normal Conquest, Arena, and Joust until they reach account level 5, at which point all gamemodes become available for play. In addition to PvP, PvE versions of these modes are also available, allowing players to team up against AI-controlled bots for a more casual or practice-oriented experience.

Conquest
Conquest is a strategic 5v5 battle on a large map with three lanes, jungle camps, and major objectives. As SMITE 2''s premier mode, it emphasizes teamwork, strategy, and objective control, making it the go-to mode for players looking to engage in traditional MOBA gameplay. In addition to Normal Conquest, a Ranked variant is available, featuring a Draft Pick system where players take turns banning and selecting gods.

Joust
Joust is a high-stakes 3v3 mode played on a single-lane map, emphasizing team skirmishes and objective control in a tighter, more intense setting. A 1v1 variant, called Duel, is also available for players who want to prove themselves in solo combat, where individual skill and tactical decision-making are key to victory.

Assault
Assault is a chaotic 5v5 mode set in a single lane where players are randomly assigned a god, creating unpredictable, fast-paced battles, and emphasizes adaptability, which rewards players who can make the most of unexpected god selections.

Random Gods: Players are given a random god at the start of the match but have the option to swap with teammates or re-roll for a different one.

Practice
Practice mode is a casual environment designed for players to experiment with abilities, items, and game mechanics without any pressure or competitive objectives. While not considered a “real” gamemode, it is an essential tool for learning and refining skills, making it ideal for both newcomers and experienced players looking to optimize their gameplay.
